Naturally aspirated V12 in a rear mid-engine position, enlarged to 5.2 liters on the latest versions. Rear-wheel drive, manual gearbox, tubular chassis, and aluminum body. The 2021 Countach LPI 800-4 reinterpretation reuses the hybrid V12 architecture of the Sián.

Les Lamborghini chaussent des pneus de très grand diamètre et à profil bas, avec montage étagé (pneus arrière plus larges que l'avant) sur les modèles sport. L'Urus utilise couramment des jantes de 21, 22 et 23 pouces. Les voitures sport comme la Huracán emploient des dimensions ultra-performance en 19-20 pouces. L'entraxe (bolt pattern) le plus fréquent est en 5 trous, partagé avec d'autres modèles du groupe Volkswagen. Au Québec, le passage à des pneus d'hiver homologués est essentiel et obligatoire : il faut privilégier des dimensions hiver adaptées, souvent sur jantes de diamètre inférieur, pour préserver l'adhérence par temps froid.
